No you can't. If you're referring to the split screen co-op mode, then it's been taken off on the PC version. Only console versions had that feature.
I want to know whether I can play dual player offline on a single PC with Controller and Keyboard. Since RE5 has two characters, I want that one to be operated by me through Keyboard and other to be operator by my friend on controller. In my case, both controller and keyboard controls one character at the same time and the other character remains auto controlled by the computer. Please help in this regard. I am using xbox 360 controller for PC and normal qwerty keyboard for gaming.

I did play offline co-op with a friend over a LAN connection.
